Bendigo Police confirm a 28-year-old Epsom man is one of two people who lost their lives in a multi-vehicle collision in Harcourt North on Friday night.
A 35-year-old Ballarat woman also lost her life when four cars and a truck collided on the Calder Freeway near Fogartys Gap Road just after 9pm.
There are reports a vehicle was driving southbound in the northbound lanes shortly before the crash.
Bendigo Police Highway Patrol leading senior constable David Hockey says that one of the vehicles involved in the multi-vehicle incident is being investigated for driving south in the northbound lanes.
"We are seeking witnesses in relation to a vehicle being seen driving in the northbound lanes when travelling south," Mr Hockey said.
Both fatalities were drivers of single occupant vehicles.
"We are seeking any information that can be related to the incident prior to it actually happening," Mr Hockey said.
"In particular, anyone who has seen a vehicle travelling from the Big Hill area south in the northbound lanes."
Three people were assessed at the scene for injuries, with two people taken to Bendigo Hospital.
Mr Hockey said the identity of the deceased would not be confirmed until the Coroner's report is completed.

An Ambulance Victoria spokesperson said it received a call at 9.11pm on Friday night to attend a multi vehicle collision in Harcourt North.
The spokesperson says three people were assessed at the scene, with two people taken to Bendigo Hospital.
A man, believed to be in his 50s, suffered a shoulder injury and was transferred to hospital in a stable condition.
A woman, believed to be in her 20s, suffered a chest injury and was taken to Bendigo Hospital in a stable condition.
Another man, believed to be in his 60s, was injured but did not need to go to hospital.
Ambulance Victoria did not provide information about the two deceased persons.

Police are investigating after a fatal multi vehicle collision in Harcourt North last night.
Two people, who have yet to be formally identified, died at the scene and three were taken to hospital with non-life-threatening injuries.
It is believed four cars and a truck collided on the Calder Freeway near Fogartys Gap Road just after 9pm.
The exact cause of the collision is yet to be determined.
