Police said two people, who had yet to be formally identified, died at the scene.
Three people were taken to hospital with non-life-threatening injuries.
Police have yet to determine the exact cause of the crash, but said it was believed four cars and a truck collided on the Calder Freeway near Fogartys Gap Road shortly after 9pm.
The fatal crash brings the state's provisional road toll to 72, which is five fewer lives lost than this time last year.
