The film - Road to Boston - will shoot in parts of Pall Mall, Park Road, and Wattle and Mackenzie streets over a number of days from January 17 to 21.
The film is based on the true story of a Korean marathon runner who competed in the 1947 Boston Marathon.
Extra roles include crowd members, journalists, marathon staff, and marathon runners.
No acting experience is necessary - cast members only need to be available for a costume fitting in Melbourne later this month and for filming in Bendigo in late-January.
Geelong, Ballarat and Melbourne will also feature in the film's production.
Anyone interested in auditioning should head to the Engine Room on View Street between 3pm and 7pm on Monday, December 16.
