Firefighters, paramedics and police were called to the intersection of the Calder Freeway and Pollards Road about 6pm.
Paramedics took one patient to Bendigo hospital in a stable condition.
Police believed the woman, aged in her 30s, had sustained an arm injury.
Castlemaine Sergeant Robert Gibson said it appeared one party had failed to give way, causing the crash.
He said one vehicle was attempting to cross the intersection. The other vehicle was travelling along the freeway, in a section with a speed limit of 110 kilometres an hour.
Sergeant Gibson said one car clipped the rear of the other.
There were two children in one of the vehicles, and one child in another.
Sergeant Gibson said paramedics assessed all three children at the scene.
The injured woman was the only person taken to hospital.
Police investigations into the crash are ongoing.
Sergeant Gibson urged anyone with information to call the Castlemaine Police Station on 5470 4100 or to make an anonymous report to Crime Stoppers on 1800 333 000.
