Bendigo Highway Patrol Senior Sergeant Ian Brooks said Lockwood Road is likely to be closed for another hour while police continue their investigations into the cause of the crash.
"It is very early in the investigation. We don't have a lot of information," he said. "We do have some witnesses who are helping with the investigation but it is too early in any part of the investigation to say what actually happened.
"At this stage we think he has drifted off the road but its too early to make judgments. Hopefully by this time tomorrow we will have a definitive answer."
Senior Sergeant Brooks said emergency services were called to the crash scene just after 3.30pm after reports of a car crashing into a tree.
"(Emergency services) responded and removed a 23-year-old male from the car who, at that time, was still alive but struggling," he said. "Ambulance services tried their hardest to keep him alive but, unfortunately, he died at the scene."
"Some passers by generously stopped and put themselves in a position they probably didn't want to be in to try and save this man's life."
Senior Sergeant Brooks urged drivers to take care on the roads as traffic increased due to coronavirus restriction being lifted.
"At this time of year we are getting a lot more traffic on the roads and are expecting more from next week," he said. "We ask people take care and drive to the conditions."
Police will prepare a report for the coroner. So far 95 lives have been lost on Victorian roads this year compared to 126 at the same time in 2019.

THE MAN who died this afternoon is a 23-year-old from Bendigo, police say.
Paramedics treated the man but he died at the scene.
4.45pm
ONE person is dead following the crash in Lockwood.
One maroon Mazda sedan has been extensively damaged in the crash. The car has hit a tree on the side of the road.
Police have blocked Lockwood Road in both directions. Traffic diversions have been set up at the Calder Alternative Highway and Wiegard Road.
VicRoads is urging people to follow the directions of police at the scene and to use the Calder Freeway or the Calder Alternative if travelling in the area.
4.30pm
AN AIR ambulance has been dispatched from the Bendigo Hospital to the crash scene.
Paramedics are assessing one person involved in the crash.
Police are diverting traffic outbound from Bendigo down Wiegard Road.
4.15pm
A VEHICLE has crashed into a tree in Lockwood.
All emergency services are on scene.
An SES crew arrived five minutes ago in case a patient needed to be extracted from the vehicle.
Police are blocking eastbound traffic along Lockwood Road.
4pm
EMERGENCY crews are en route to a reported car crash on Lockwood Road.
VicEmergency is reporting the crash 500 metres from the Calder Alternate Highway at the McCombs Road intersection.
Paramedics are on scene assessing the situation.
Firefighters and police have also arrived.
