The 6.44pm Southern Cross to Kyneton service will not run today after a carriage was vandalised overnight.
Passengers are being advised to consider the 7.02pm service from Southern Cross to Bendigo.
Fumes from spray paint used to graffiti the exterior of a carriage leaked into the interior of the train, rendering it unfit for service.
It resulted in the 5.13am service from Kyneton to Southern Cross being cancelled with the train being removed from service to be cleaned, causing a short-term reduction in services.
Due to lack of available coaches at that time of the day, passengers were asked to board the next available service which departed at 5.48am.
"This illegal behaviour has a real impact on our passengers, whether they are trying to get to work on time or connect with other transport services in Melbourne," a V/Line spokesperson said.
"The incident has been reported to Victoria Police and we will be helping with their investigation."
Anyone with information is encouraged to call Crimestoppers on 1800 333 000.
