The top eight teams in each division qualified for the knockout finals series, which will be played over the next three weekends.
The Super Cup is a seeded knockout event, with Sunday's opening round featuring first versus eighth, second vesus seventh, third versus sixth and fourth versus fifth.
Bendigo venues Epsom, Shadforth Park, Stanley Avenue and Truscott Reserve will host Sunday's matches.
The semi-finals on September 29 will be played in Shepparton, with the grand finals on October 6 at Beischer Park in Strathdale.
The top two seeds in division one - Strathdale and Eaglehawk - are expected to have few troubles in getting past Colts United and Golden City respectively.
The other two matches are a different story.
Third-seed Moama-Echuca and fourth-seed Tatura arguably go into their respective games with Shepparton South and Shepparton United as underdogs.
Shepparton South has scored 27 goals in its past four games, including a 6-0 drubbing of Moama-Echuca less than a month ago.
Shepparton United is the only team to defeat Strathdale all season - a 1-0 result last weekend.
It's a similar story in the opening round of the women's Super Cup.
Championship-winners Shepparton United and runner-up Eaglehawk should stroll through their opening matches against Golden City and Spring Gully respectively.
However, the remaining two games coud go either way.
The fourth versus fifth battle between Shepparton South and Strathdale will be a tight affair, while sixth-seed La Trobe University is in season-best form ahead of its battle with third-seed Colts United.

Division one (all games at 3pm) - Strathdale (1) v Colts United (8) at Epsom, Eaglehawk (2) v Golden City (7) at Truscott Reserve, Moama-Echuca (3) v Shepparton South (6) at Shadforth Park, Tatura (4) v Shepparton United (5) at Stanley Avenue.
Women's division (all games at 10.30am) - Shepparton United (1) v Golden City (8) at Stanley Avenue, Eaglehawk (2) v Spring Gully (7) at Truscott Reserve, Colts United (3) v La Trobe Uni (6) at Epsom, Shepparton South (4) v Strathdale (5) at Shadforth Park.
