The extension will add an extra 4000 square metres of floor space to the store, create more jobs and add an extra 60 spaces to the car park.
The project will take the floor space of the store - which has now been in Bendigo for 20 years, to 13,500 square metres.
By comparison, the new store at Delacombe in Ballarat is one of the largest stores in Australia at 17,315 square metres.
Bunnings says the project will lead to a larger main warehouse, as well as a larger outdoor nursery, timber section and building materials landscape section.
"We're expanding the Kangaroo Flat store in order to offer the Bendigo community an improved and expanded store which will allow us to stock an even wider range of products," complex manager Mark Galvin said.
The store is expanding to a site that sits adjacent to Railway Street.
The company flagged an expansion to the store back in 2014.
Work is expected to finish by Easter. The shop will continue trading throughout.
