Kalamazoo Resources have started scouring the 93 square kilometre stretch of land south-east of Maldon after securing an exploration license, the company said in a statement to the ASX on Wednesday.
The Maldon goldfield has produced over two million ounces of gold since the dawn of mining in the region.
Kalamazoo's new exploration plot is considered "under-explored" with only minor exploration in the past 10 years, the statement read.
"No systematic modern exploration has occurred in this area, e.g. geophysics surveys."
The new "South Muckleford" site sits 10 kilometres east of Kalamazoo's Wattle Gully and Wattle Gully exploration zones, which stretch south from Castlemaine.
The new site is mostly on crown land.

Modern, "low impact" ground surveys will be used in areas Kalamazoo believes could be most valuable, the company said a statement to the ASX on Wednesday.
"This is in keeping with the company's philosophy of conducting exploitation in a 'smarter', low impact, reconnaissance-style methodology that minimises any disturbances to the environment and the local community,"
"They essentially reduce the amount of ground disturbance exploration drilling required to effectively explore a prospective area."
