Emergency services were called to the fire, in Havelock Street, Dunolly, about 12.55am.
The vehicle, which was completely destroyed, had an off-road camper trailer attached. The trailer was stolen prior to the vehicle being set alight.
Goldfields Criminal Investigation Unit members are investigating the car fire and trailer theft.
The cause of the blaze remains unclear, but investigators believe the blaze was deliberately lit.
Fire crews stayed on scene until 2.17am.
Police asked anyone who might have seen a person or persons acting suspiciously in the area around the time of the fire to come forward.
In particular, they are interested in speaking to anyone who might have seen the stolen trailer.
Investigators have released pictures of the stolen trailer in the hope that someone knows its whereabouts.
The blaze follows another suspicious car fire earlier this week.
On Monday, Bendigo police and Country Fire Authority crews were called to a car fire in Fenton Street, Bendigo, just before midnight.
A blue Ford sedan had been set alight.
Police asked anyone with information on the fires or theft to call Crime Stoppers on 1300 333 000 or submit a report online via crimestoppers.com.au.
