About 1.40am Friday, a Holden Commodore ute with allegedly stolen number plates was seen driving along the Princes Highway at Laverton.
Police monitored the vehicle, which allegedly began speeding up and driving erratically on Old Geelong Road at Hoppers Crossing.
The same vehicle was later seen entering the car park of an entertainment complex at Southbank.
The driver fled the ute, but was chased by officers and arrested.
The 28-year-old from Sidonia, near Kyneton, was taken to hospital under police guard and will be interviewed later.
Police also allege a quantity of drugs was found in the car.
A constable was allegedly struck during the arrest, but she sustained no injuries.
