Emergency services were called to an Elizabeth Street address just before 8.30pm following reports of a house fire.
The house was well alight when firefighters arrived, according to a CFA spokesperson.
The fire was brought under control within about 25 minutes, and has since been declared safe.
Four CFA appliances responded, the majority of whom were from Echuca.
The CFA spokesperson said three people were in the house at the time of the fire, all of whom self-evacuated.
An Ambulance Victoria spokesperson said a woman in her 40s was taken to Goulburn Valley Health in a stable condition.
Another woman, in her 20s, was taken to Echuca Regional Health following assessment for mild smoke inhalation.
A third person, a high school-aged girl, was also assessed for smoke inhalation. However, the Ambulance Victoria spokesperson said transportation to hospital was not required.
The house was extensively damaged by fire, according to the CFA spokesperson.
He said the fire was not believed to be suspicious in nature, but that fire investigators were expected to attend in the morning.
The cause of the blaze is not yet known.
Firefighters have attended a number of house fires throughout the region in recent weeks, including a fire at an historic hill station on Mount Macedon on Wednesday.
An historic homestead in Kyneton, owned by TV personality Tonia Todman, was significantly damaged by fire on Monday.
Mount Macedon Volunteer Fire Brigade members urged people on Wednesday to take precautions against fires in the home, such as fitting fireplace guards and smoke detectors.
