The change will affect an area bordered by Barnard, Myrtle, View and Chapel streets, and the railway to the south.
The main arterial roads in the precinct – Wills, Myers, Myrtle, Chapel and High streets – will not be affected by the new speed limit.
The change is part of a suite of measures designed to make the CBD more pedestrian-friendly.
Other changes include alterations to existing roundabouts and 18 new zebra crossings, including one in View Street near the Bendigo Art Gallery.
Transport Accident Commission modelling has estimated the changes would see a 30 per cent reduction in crashes in the CBD.
