The northbound lanes of High Street will be widened and the right turn lane on to Myrtle Street extended to prevent congestion.
Currently, the queue of vehicles waiting to turn right often extends beyond the turning lane in busy times, blocking a lane of traffic attempting to travel straight through.
VicRoads regional director Brian Westley said 20,000 vehicles travelled High Street each day, while 13,000 used Myrtle Street and 15,000 used Don Street.
The project will also include a new bicycle lane on High Street between Don Street and Violet Street, the resurfacing of the asphalt pathway for pedestrians, and a new pedestrian island on Violet Street.
Mr Westley said the presence of a tourist attraction, Central Deborah Gold Mine, at Violet Street meant there would be people trying to cross the road.
He said it significantly reduced the risk to the pedestrians when the distance of road they had to cross was reduced.
With the upgrade to take place on such a busy road, Bendigo West MP Maree Edwards said there would be efforts to undertake work at times of least disruption to motorists, such as at night.
The work is scheduled to finish by the end of June.
