The burn, between Mitiamo and Prairie, will likely start between 9.30am and 10am.
Motorists will have to take detours, with signs to be put in place to advise of suitable routes.
While the CFA and Forest Fire Management Victoria have begun planned burns, the fire danger period remains in force across some parts of the region.
The municipalities of Central Goldfields, Greater Bendigo, Macedon Ranges and Mount Alexander remain subject to fire danger period restrictions until May 1.
During the fire danger period, burn-offs are banned without a permit from the municipal fire officer, CFA or the fire prevention officer of a public authority.
Restrictions also apply to other activities, including the lighting of camp fires, the use of incinerators and hot work.
