The offenders gained entry to the building by smashing a glass panel in the door of the Caltex service station in Johnstone Street.
They stole the till and a box of cigarette lighters.
The till was later discovered on the side of the road on the Midland Highway in Barkers Creek, but about $300 in cash had been stolen.
Police also found some cigarette lighters scattered at the front of the service station.
No suspects have yet been identified, but officers are reviewing CCTV footage.
Anyone who might have witnessed the incident or has any information is asked to contact Castlemaine police station on 5470 4100.
EARLIER: Investigations will continue on Tuesday into a burglary at a Castlemaine service station.
The crime at the Caltex service station in Johnstone Street was reported to police shortly before 2am.
Attending officers found damage to the front door and various items strewn outside.
At this stage, police are unable to say what might have been stolen.
