Bendigo Sergeant Mark Holloway said a caravan, which one of the vehicles was towing, ended up on its roof.
Emergency services were called to the Bendigo-Maryborough Road at the Newbridge turnoff just before 5.30pm and remain at the scene.
No-one is believed to have been injured.
Paramedics attended an unrelated traffic incident earlier in the afternoon near Bears Lagoon – the second in two days.
A woman was transported to Bendigo Health in a stable condition after a car crashed into a pole on the Loddon Valley Highway about 3.30pm on Friday, an Ambulance Victoria spokeswoman said.
The highway was the site of a multi-vehicle crash on Thursday afternoon.
No-one was injured.
