Bendigo got into the spirit of the occasion with 32 registered events on the day, with a host of other private events to help raise money for the RSPCA.
McIvor Road Veterinary Centre was among those to have the frosted delights available throughout the day.
Administration staff member Anna Pinton said the clinic was happy to support the RSPCA.
"We do de-sexing and look after a lot of their animals," she said.
"One of our staff members made some lovely cupcakes and they were very nicely decorated."
The centre dished out plate after plate of animal-themed cakes to staff and clients throughout the day.
Described as "the sweetest way to fight animal cruelty", the annual day raised $1.2 million across Australia in 2013.
RSPCA spokesperson Caitlin Barratt said because the organisation receives less than three per cent government funding, days such as Cupcake Day were vital to RSPCA Victoria.
She said the RSPCA relied heavily on the "kindness and generosity" of the public to allow it to continue its animal welfare work.
