The Australians scored 200.26 after shooting at the 300m and 500m targets.
They are just behind the English pair on 200.29, with Jersey in third place on 199.26.
The shoot-off rolls on Saturday at the 600m, 900-yard and 1000-yard distances.
This is Grenfell's third Commonwealth Games campaign.
He won gold in the full-bore pairs event at the 1994 Commonwealth Games in Victoria, Canada, and competed in Delhi four years ago.
The shooting champion has won the Bendigo Advertiser-Prime TV Sports Star of the Year award and was inducted to awards Hall of Fame last March.
