Police said the woman, 64, was crossing the road near The Good Guys shop in Williamson Street just after midday when a grey Holden Commodore pulled out of a park in the centre of the road.
The car struck the woman, running over her legs.
Police said the man was believed to be looking left for oncoming traffic when he pulled out of the car park.
An Ambulance Victoria spokesperson said the woman was taken to Bendigo Health with multiple injuries.
Victoria Police later released a statement advising that the woman had died shortly after being admitted.
Bendigo Senior Constable Dave Sanders said police were called to the scene on Williamson Street at 12.19pm.
He said the car hit the woman at a slow speed and alcohol was not a contributing factor.
Senior Constable Sanders said drivers needed to be mindful of either direction when leaving from a parked position.
"You have to look both ways moving out of car parks, especially in town," he said.
The male driver and his passenger, of Marong, were uninjured in the incident.
Senior Constable Sanders said the driver was assisting police with their inquiries.
He said the matter would be dealt with by a Coroner in due course.
