Police allege a truck heading south along Woodward Road collided with a cyclist at the intersection of Hattam Street about 12.10pm.
The cyclist died at the scene.
A 42-year-old Quarry Hill man has been charged with dangerous driving causing death.
He faced an out of sessions hearing and has been remanded to appear at Melbourne Magistrates' Court on Tuesday.
Update, 5.30pm: AN 18-year-old man has died after the bike he was riding was hit by a truck at Golden Square.
Major Collision Investigation Unit detectives believe the truck, which was travelling towards Hattam Street along Woodward Road, went through a give way sign.
The cyclist was travelling towards Bendigo on Hattam Street when he was hit.
Police have yet to determine why the crash occurred.
The truck driver, a 42-year-old Quarry Hill man, is assisting police with their inquiries.
Investigators expect to remain at the scene for about another hour.
The affected sections of Hattam Street and Woodward Road remain closed. Detours are in place.
The 18-year-old man was the 145th person killed on Victorian roads this year.
EARLIER: EMERGENCY services are on scene of a fatal collision in Golden Square.
Police understood a truck heading south along Woodward Road collided with a cyclist at the intersection of Hattam Street about 12.10pm on Friday.
The male cyclist, who is yet to be formally identified, died at the scene.
The male truck driver stopped at the scene and is helping police with their enquiries.
Police are yet to establish the exact circumstances surrounding the incident and the investigation is ongoing.
Major Collision Investigation Unit detectives are also on their way.
Road closures are in place along Hattam Street from Belle Vue Road and Vains Street through to MacDougall Road.
They are expected to remain for several hours.
