The busy intersection of Brunswick Street and Alexandra Parade on the border of Fitzroy and Fitzroy North was closed after a Mercedes Benz collided with a motorcycle at about 10.45am on Friday.
Ambulance Victoria said the motorcyclist was rushed to the Royal Melbourne Hospital in a serious condition with head injuries.
On Saturday morning he remained in a critical condition, a hospital spokeswoman said.
Police said the 41-year-old male rider from Ringwood had life-threatening injuries while the 61-year-old male driver was uninjured.
The cause of the crash is being investigated and police are appealing for witnesses or dash-cam footage.
Traffic ground to a halt causing havoc in the area, and trams were blocked from travelling up Brunswick Road through Fitzroy.
The intersection has re-opened and tram routes on Brunswick Road are operating as normal.
Route 11 trams have resumed along normal lines after an earlier disruption. Delays may occur as we restore the timetable.??? Yarra Trams (@yarratrams) January 19, 2018
