It is believed the 60-year-old man, from Lockwood South, was travelling south along the Calder Highway about 9.30am when the collision occurred.
He was thrown from his bike and died at the scene.
Police remain at the scene of the fatal crash and will prepare a report for the coroner.
VicRoads’ traffic website indicates southbound lanes are again open at Taradale. Cars were earlier diverted onto the Pyrenees Highway.
The man is the 201st person to die on Victorian roads in 2017.
