VicRoads said traffic will be temporarily stopped for up to 15 minutes in both directions on the Calder Highway.
The traffic stop is for the safety of drivers and road workers while construction continues to build the new Ravenswood Interchange in the centre median.
Traffic controllers will also temporarily stop Melbourne-bound traffic on the Calder Alternative Highway.
Local access to Beilharz, Bickfords and Soldier Roads will also be temporarily stopped.
Drivers should plan ahead and allow extra time if travelling in the area or to Melbourne or Bendigo between these times.
The completed interchange will be an elevated circular roadway that will separate freight and slow-moving traffic from faster moving traffic.
Work on the $86 million Ravenswood interchange began last year. It is expected to be completed in late 2017.
