Entering the last stage of the competition, the 3200m run and pistol shooting, the 24-year-old was placed seventh, starting 45 seconds behind the leader.
However, she worked her way quickly through the field and with one of the four laps remaining took the lead from Poland's Oktawia Nowacka when she landed five targets quickly.
Esposito then strode out to win by 16 seconds. She broke down as she embraced her coach and father Daniel, an Olympian in the sport in 1984.
Australia's Chef de Mission, Kitty Chiller, herself a modern pentathlete who represented Australia at the Sydney Olympics in 2000, was at the venue celebrating Esposito's success.
The modern pentathlon comprises five different events: fencing, 200m freestyle swimming, show jumping, and a final combined event of a 3200m run and pistol shooting.
