A source at the jail in Castlemaine told Fairfax Media one of the guards involved in Thursday morning's alleged scuffle had lost six front teeth, had his nose broken and received a "significant gash" to his face.
The other guard sustained minor injuries.
The prison officers approached the inmate to conduct an "enter and extract" after he refused to leave his cell, according to the source.
Victoria Police are on the scene.
The facility houses around 200 inmates.
The facility was in lockdown but, according to radio station 3AW, those restrictions have been lifted.
