Played in wet and cold conditions on Thursday, Bilkey won an emotional 18-hole match play final against her 19-year-old daughter Kristi Bilkey.
Reigning champion Kristi got away to a flyer, birdying the first hole.
Jenni replied with a birdie of her own on the second, then claimed the lead after she parred the third hole.
She was never headed and finished one-up to score her fourth A-grade championship and first since 2012.
It was the first time the mother and daughter had met in the club championship final.
Jenni said it was tough coming up against her daughter in the final.
"I tried not to think too much about it leading into it, but it got to me on the 18th," she said.
"Kristi and I have tried to play each other before but we have never made it to the grand final together.
"We've made it to the semi and then she knocked me out. To met her in the final, as a parent, it's awesome.
"We are really proud of Kristi and her golf."
The mother and daughter embraced after Jenni sunk her putt on the 18th to ensure her victory.
"It was hard playing, because David, my husband, was caddying for Kristi," she said.
"I had to detach myself from them while I played. Leanne was my caddy and she was awesome.
"It's really satisfying winning - you need to be really focused."
The Bilkeys qualified for the final after two stroke rounds and wins in their semi-finals.
Jenni, who has been playing at the club for 32 years, previously won The Bendigo Advertiser Cup in 2005, 2009 and 2012, while Kristi won her club championship as an 18-year-old last year.
Lyn Harding won the Mrs Townsend Trophy as B-grade champion, defeating Yvonne McDonald two-up, with one hole to play.
The C-grade Mrs Douglass Trophy was won by Angela Dudley, who finished one up against Michelle Elliott.
