The 4WD travelled up to 30 metres on its side after an incorrectly loaded trailer swung around, causing the vehicle to roll about 3.30pm on Saturday.
Investigators believe the trailer, which contained another smaller trailer and fibre glass boat, turned 45 degrees before its connection with the car broke.
The two men were treated at the scene by paramedics for minor cuts and bruises before being taken to Bendigo Hospital for observation.
Melbourne-bound traffic was blocked from Furness Street, Kangaroo Flat, while SES crews worked to clear the road of debris.
Leading Senior Constable Stuart Buchanan said the driver and his passenger were lucky to have avoided serious injury.
He said while trailer-related accidents were not uncommon, it was rare that it would cause the vehicle to roll over.
“In this case, the nature of the vehicle, being a 4WD, has caused it to roll,” he said.
No other vehicles were nearby at the time of the crash.
Leading Senior Constable Buchanan reminded drivers to ensure trailer loads were balanced, with weight on the tow bar, to prevent such incidents from occurring.
