Griffin earned nine votes to win the power forward position ahead of the Townsville Fire's Cayla George (six votes).
Alaskan-born Griffin has represented the Spirit for four seasons, which have included two championship wins.
The two-time WNBL grand final most valuable player was just one of three players to average a double-double with 17.9 points and 10 rebounds a game.
She finished 10th in the league for blocks with 20 total and fourth for steals with 39.
Her All-Star nomination follows her first inclusion in the 2014/15 team.
Griffin is joined in the All-Star Five by Adelaide Lightning point guard Leilani Mitchell (10 votes), Perth Lynx shooting guard Sami Whitcomb (15), Sydney Uni Flames small forward Katie-Rae Ebzery (9) and centre Suzy Batkovic (15).
Other notable vote getters South East Queensland Star guard Lauren Mansfield (7), Perth guard Betnijah Laney (6) and Dandenong Rangers guard Steph Cumming (6).
Griffin was the Spirit's 2015-16 season MVP.
