Michael Kelly, who lives interstate, took custody of his sister's four children after she was reported missing on February 4.
The last confirmed sighting of Ms Kelly was on January 20 when she was seen in the shopping precinct in High Street, Kangaroo Flat.
Homicide squad detectives arrived in Bendigo on Monday to piece together clues as to what had happened to her since.
Mr Kelly said it was out of character for the 39-year-old to leave her children.
"She didn't have any other real interests apart from her kids," he said.
"Anything she's ever got up and done has been for those children."
He asked anyone with knowledge about the disappearance to phone police immediately.
"Report everything you can and give it back to the police so they can help find my sister - for her own sake," he said.
Despite police fears of foul play, Mr Kelly holds hope his sister might be found unharmed.
"Come home, Sammy, or let us know you're safe."
"We need you to get the help you need to take control of your kids."
Anyone with information should phone Crime Stoppers on 1800 333 000.
