The Noble Park man was trapped in the car for about 45 minutes while emergency services crews worked to free him.
He was the sole occupant of the vehicle.
Police said the 24-year-old driver was travelling southbound on the Calder Freeway before doing a u-turn and colliding with the pylon at the Malmsbury East Road overpass.
It is believed the crash was intentional.
The collision occurred at about 12.30pm.
The northbound lanes of the Calder Freeway were shut for some time as emergency services attended to the crash.
An Ambulance Victoria spokesperson said the man suffered serious leg injuries and possible pelvic injuries in the smash.
An air ambulance transported him to hospital at about 2pm.
The crash was attended by police, ambulance, SES and CFA crews.
The driver is reported to be in a serious but stable condition.
UPDATE 1.30pm: The man injured in a single vehicle crash on the Calder Freeway near Malmsbury will soon be airlifted to hospital.
An Ambulance Victoria spokesperson said the man suffered serious chest injuries when his car smashed into a pylon near the Malmsbury East Road overpass.
He also sustained leg injuries and possible spinal injuries.
It is believed he was the sole occupant of the vehicle.
UPDATE 1pm: A man believed to be in his 30s has been pulled from the wrecked car.
A CFA spokeswoman said the CFA were called to the scene at 12.34pm.
She said the occupant of the vehicle was trapped in the wreckage for a short time and is now in the care of paramedics.
An air ambulance is on scene.
EARLIER: EMERGENCY services are en route to a single vehicle crash near Malmsbury on the Calder Freeway.
Two CFA trucks are on scene with another three on the way. The SES is also travelling to the scene.
Traffic is being diverted away from the accident scene.
