Woodend Police Senior Constable Darren Kyritsis said the offending truck driver, a 50-year-old man, failed to notice a slow-moving truck in front of him and hit the back of the vehicle.
The truck in front was an observation vehicle for line-marking operations, driven by a 34 year old man.
The observation truck was carrying the line-marking paint, and has signage directing drivers to use the right lane.
The trucks collided at about 11.30am in the left hand lane, northbound, just after the Woodend/Lancefield exit.
The driver of the offending vehicle was taken by ambulance to the Royal Melbourne Hospital for observation but was released shortly after admission. His male passenger was uninjured, as was the other truck driver.
Traffic is being diverted through Woodend.
A specialist water pressure machine from Moe has been called to the scene to help with the clean up. It is expected to arrive at 6pm and work until about 11pm. Both lanes will reopen following the clean up, Sen. Const. Kyritis advised.
Sen Const. Kyritis said police are investigating the cause.

IT appears a truck has collided with the rear of vehicle protecting a line marking truck on the Calder Freeway at Woodend, a witness says.
Nino Grgurovic - who arrived about a minute after the collision - said traffic was banked up for about four kilometres on the northbound side of the road.
He said there was white paint all over the freeway.

A Twitter user has posted a photo of white paint on the Calder Freeway after a collision between two trucks - one of which was marking lines.
All northbound lanes remain closed near Romsey Road.

VicRoads is advising motorists to seek an alternate route. Emergency services are still on scene.
UPDATE, 12.07pm:
THE incident on the Calder Freeway at Woodend is believed to be a collision between two trucks.
An Ambulance Victoria media spokeswoman said two men were being treated by paramedics at the scene.
She said the men, one aged in his 30s and the other in his 50s, are both in a stable condition.

FIRE crews are at the scene of an incident on the Calder Freeway at Woodend.
The incident was reported about 11.45am on Tuesday.
