A silver medallist to Bendigo star Peta Mullens at last month's Cycling Australia road championship race at Ballarat, Neylan continued her remarkable return.
A solo attack at 8km to go paid off as Neylan won the Subaru National Road Series event along the Great Ocean Road and Bellarine Peninsula.
Runner-up was Santos Tour Down Under winner Valentina Scandolara ahead of Tessa Fabry.
“I’ve been waiting a long time to win a big race and it’s finally happened,” said Neylan who won silver at the 2012 UCI Road World Championships.
A field of 67 competed on Saturday.
Before the women's event more than 3500 recreational cyclists took to the road in the Momentum Energy People’s Ride, led by Prime Minister Tony Abbott who rode the 65km course.
