The petition, supported by community group Axedale Our Town - Our Future, would put the voice of the community behind the CFA's planning amendment submission to bring the siren back online.
The CFA was in the process of formally requesting to bring the siren back with the City of Greater Bendigo.
Axedale Our Town - Our Future's monthly meeting on Wednesday attracted almost 70 people, most of which were keen to see progress on the siren issue.
Committee president Yvonne Wrigglesworth said the issue had been one of the biggest in the community in recent history.
She said the committee made the decision as a group to support a petition.
"The petition will guarantee there will be strong support in favour of the siren," Ms Wrigglesworth said.
"We are aiming to gather 500 signatures by Sunday night, which I think we will be able to achieve.
"So far we have been receiving support from far and wide."
A representative from the region's CFA and the City of Greater Bendigo attended Axedale Our Town - Our Future's monthly meeting.
Ms Wrigglesworth said they explained the situation clearly to those in attendance.
The town of just over 600 people was told it could no longer operate the siren at its fire station from last Wednesday.
Axedale's new fire station was opened earlier this year, but two objections regarding the siren meant the CFA did not have planning approval to operate one from the site.
The CFA was expected to submit its application by the end of the month, with the council to display to the public for two weeks. People would then have an opportunity to object to the siren.
Ms Wrigglesworth said the petition would continue to circulate for another two weeks before it was used to support the CFA's planning amendment.
