Flood waters inundated the former hospital in January 2011, causing significant damage and left the town requiring a complete rebuild of its main health service.
The new hospital includes acute beds, residential aged care beds, urgent care and GP consulting rooms. Flood resilience measures were also included in the design.
Minister for Health David Davis opened the $22.7 million hospital, which formed part of the East Wimmera Health Service.
Mr Davis said the new hospital was a reflection of the resilience of the Charlton community to recover from the natural disaster.
"The modern new hospital will see the return of vital health and aged care services to Charlton after the former hospital sustained irreparable damage in the floods," he said.
