There will be two daily return services from Epsom to Southern Cross in Melbourne.
On Monday to Friday, Epsom to Southern Cross services will leave at 5.58am and 8.25am.
Return services on weekdays will leave Southern Cross at 3.15pm and 5.31pm.
On Saturday and Sunday there will be one morning and afternoon train from Epsom to Southern Cross.
Epsom Railway Station has 100 free car spaces, a drop-off zone and bike racks. Bus route 10 will be altered to operate via the station.
An extra morning service to Melbourne will be added to Eaglehawk Railway Station from Monday October 13.
Myki transport cards are required for all journeys between Epsom and Southern Cross, unless travellers have a V/Line family traveller ticket.
Transport minister Terry Mulder said the state government rebuilt Epsom Railways Station from scratch.
"It has been 40 years since passenger trains stopped at Epsom," Mr Mulder said.
Residents are invited to a public event from 10am at the station on Saturday October 11 to celebrate its opening.
