The record attempt was part of a fundraiser for the Ride to Conquer Cancer with all money going to the Peter MacCallum Cancer Centre.
Organiser Lisa Nicolson said the group fell short of achieving the record of 498 - set on Bondi earlier this year - but said about $8000 was raised nonetheless.
"We had about 100 packs left over, so people have registered but not turned up which is disappointing because we would have killed it otherwise," she said.
Despite not breaking the record, Ms Nicolson said she was overwhelmed with the turnout.
"I am pleased with how many people turned up. It was pretty cool seeing a sea of black shirts and coloured gloves," she said.
Ms Nicolson said the event aimed to bring together people who had in some way been touched by cancer.
She said she knew many people who had beaten or were living with cancer, including her mother - a survivor.
"The reason why I did the fundraiser was because we need to fund raise $12,500 to enter (the ride), between the five of us so we thought we would try and do something a bit different," she said.
The Ride to Conquer Cancer is on in October.
Ms Nicolson said the Peter MacCallum Cancer Centre was an important resource for cancer patients.
"I will never forget my first time there, I specifically remember seeing a dad with his little girl in a wheelchair," she said.
"The support that the staff and the doctors have, it's constant contact - mum still visits there frequently.
"It's an amazing place and what they do."
Ms Nicolson said the group needed about $5000 to register for the ride.
