Flight Centre Park at Strathfieldsaye will host the Tab.com.au Charity Football and Netball Day which will raise money for the Children's Tumour Foundation.
Regional Victoria's four biggest harness racing areas - Bendigo, Ballarat, Melton and Shepparton - have formed football and netball teams to vie for the trophy.
The four teams will compete in a round robin series, with the grand final to be played late in the afternoon.
Bendigo won the inaugural prize last year, with former South Bendigo coach Danny O'Bree the star performer on the football field.
O'Bree is back on deck this year and Bendigo is desperate to retain the prize.
O'Bree has drafted in his former South Bendigo team-mates John Hardinge and Luke Beattie.
Bendigo's team will be coached by Larry Eastman, while Scott Dyer will be a key player.
Leading driver Ellen Tormey headlines Bendigo's netball team.
Tormey, who plays netball for Charlton in the NCFL, heads to the United States on Monday for a seven-week scholarship.
Last year's event raised $14,000 and the aim this year is to pass the $25,000 mark.
Aside from the football and netball action, the day includes a charity auction and monster raffle.
Harness racing-related businesses have thrown their support behind the day.
Alabar Stud has donated a service to star sire Grinfromeartoear for the major raffle prize.
John Campbell from Loddon Valley Stud in Lockwood has also jumped on board by donating a service to prominent stallion Union Guy.
Tabcorp has donated the uniforms for the four teams.
An auction will take place after the football and netball is completed, with sporting memorabilia and a host of goods and services up for grabs.
The football and netball starts from noon on Sunday.
Admission is $50 for players and spectators, with children under-16 free.
The $50 covers sausage sizzle, children's entertainment and entry to after game function.
