Victoria Police Media reported the incident happened about 500 metres north of the Kilmore overpass about 4.40pm.
Police believe the driver, who was travelling south on the freeway, lost control of his car, which crashed into an embankment and rolled.
The driver, a Sunbury man, was the only occupant and was ejected from the vehicle.
He died at the scene.
Police will prepare a report for the Coroner.
The incident comes after two people died and five were injured in a horror crash on the Calder Freeway in Macedon last Friday night.
