A majority of businesses along Barker and Mostyn streets showed their support to encourage people in the region to begin forming a team in the 24-hour event.
Castlemaine Relay for Life committee member Nicki Renfrey said businesses came up with some unique ideas.
"Our eventual winner 213 Hair Design had a purple-themed day, where they gave purple streaks in people's hair for a gold coin donation and had music about purple playing all day," she said.
"We had a difficult time deciding who did the best job."
The town will host the event for overnight on November 7 and 8 at Castlemaine Camp Reserve.
Ms Renfrey said it would be "fantastic" if they could draw a team each from Newstead, Harcourt and Maldon.
