Desiree Crandell created the first of the seasonal events in September 2013, called Instameets.
More than thirty artists are expected to descend on Rosalind Park on Saturday morning.
They will walk around the town, following an itinerary based on capturing the most beautiful places in Bendigo.
Ms Crandell, founder of Bendigo's seasonal Instameets, said she loved the social aspect of Instagram.
"I am not in it for numbers of likes or followers, I never have been," she said.
"It's purely about connecting and having a positive impact on people even though they may be across the other side of the world.
"It's not only a creative outlet, it has been a source of encouragement, engaging with real people and also creating real friends from sharing a common interest.
"The Instameet is an opportunity to meet new people and have face-to-face interaction.
"To laugh and share technical tips with each other as well as explore new locations and places.
"From a business point of view it's important too.
"I have visited local cafes and wineries etc, based on what I saw on other people's Instagrams."
Partnering in the event, Bendigo Tourism industry marketing and sales coordinator Mark Orlandi is preparing for an influx of creativity.
"We will have prizes available for outstanding pictures," he said.
"The event will use the social media channel that is growing so quickly.
"It will encourage people to explore and promote Bendigo."
"We will get them into the old gaol, the art gallery and the Great Stupa - all the beautiful places in Bendigo."
People are asked to hashtag #showusbendigo #explorebendigo and #autumn_in_bendi.
