The club will take on Richmond in Bendigo's second home game.
Bendigo's first hit-out on its home pitch in Barnard Street ended in a 2-2 draw with Moreland Zebras.
It's been 13 days since FC Bendigo went down 2-1 at St Albans.
Coach Steve Martin said the extended break had given the squad time to work on some areas ahead of this clash.
"It has been good for us to stop after the first four games and see where we are at," Martin said.
"It's given us the chance to go back and see where we have gone wrong in games and work on those areas."
The Easter break was a chance for some Bendigo players to rest and recover from some niggling injuries.
The only player unavailable for this fifth-round contest is Daniel Reeves, back injury.
FC Bendigo is 12th but been extremely competitive in the National Premier League division one action.
Richmond has endured a topsy-turvy start.
Coached by Grant Brebner, a former Melbourne Victory midfielder, Richmond was beaten five-nil by Kingston City last round.
Key players for Bendigo include captain Trent Waterson in forward or across midfield, Matt Breeze in midfield, and Ryan Davidson.
Waterson's pace and skill will be a major threat to Richmond.
Back playing where his career kicked off, Waterson is one of the most experienced players in the Bendigo line-up.
Waterson and Amarah Kromah scored Bendigo's goals in the clash with the Zebras.
Playing at full-back, Davidson has impressed in all four matches.
Saturday night's play kicks off a big weekend of soccer as the Bendigo Amateur Soccer League's championship begins.
The National Premier League action at the Tom Flood Sports Centre starts at 5pm when the under-20s take to the pitch.
Bendigo's under-20 team has a 1-3 record and is 10th on the ladder. Richmond is second in the championship race.
FC Bendigo and Richmond contest the main game from 7pm.
