The 43-year-old man died when a truck struck him about 7am at the intersection of Three Chain Road and Dons Road.
Police said the man was one of five contractors working at a Macedon Ranges Shire Council roadworks site and died on scene.
Macedon Ranges Crime Investigation Unit Detective Sergeant Steve Cole said police believed the truck, which was working at the site, backed into the man.
He said a number of witnesses were assisting police with the investigation.
He said police units from the Macedon Ranges, Kyneton, Gisborne and Woodend attended the accident.
The council's chief executive Dale Thornton said he was shocked and saddened by the tragedy.
"We are assisting the police and WorkSafe with their investigations, and have also launched our own investigation into the incident," he said.
"Our thoughts are with the man’s family and loved ones."
WorkSafe said it was investigating the circumstances surrounding the accident but could not comment further.
Three Chain Road was blocked in both directions for several hours yesterday.
Meanwhile a motorcyclist was killed at Bung Bong, near Maryborough, after he collided head-on with a truck about 9am Wednesday.
Police and paramedics were called to the scene of the collision at the intersection of Avoca-Talbot Road and the Pyrenees Highway.
The 46-year-old truck driver was helping police with their inquiries.
The fatalities followed the death of a truck driver in south-east Melbourne at 5.30am Wednesday.
The man, aged in his 20s, clipped a tree and power pole then went through a fence and into a parked car in Hampton East.
