Riders from under-11s through to senior age groups will contest the titles on a new course starting in High Street.
Competitors will ride anti-clockwise around Market Street, Harmony Way (the old Calder Highway) and Victoria Street, passing Stanley Park, the Harcourt Tavern and the ANA Hall.
Run by the Castlemaine Cycling Club, the championships have attracted more than 170 entrants from across Victoria.
The Showmen's and Showwomen's Criterium races will be held afterwards, with prize-money up for grabs.
