Starting off scratch and cheered on by a big home crowd, O'Shea blitzed the field on the final lap in an impressive performance to win the Golden Mile for the first time in a time of 1:38.35.
"It's the first time I've been back here for about three years, so it's always good to have that home crowd support," O'Shea said.
"This is the first time I've won the Golden Mile. I've finished second and third before, so it's good to win it.
"I thought coming into the final straight I might have gone a bit early, but I put my head down and got to the line just in time.
"This ranks right up there because it has taken me a while to win it. I love racing handicaps in Australia, so it's good that I can now tick this off."
Shane Archbold finished second and OShea's Madison team-mate Alex Edmondson was third.
