The Transplant Cricket Australia Lucky Stars will take on the DonateLife Everyday Heroes in the match at Eaglehawk's Canterbury Park on February 23 from 10am.
The game aims to raise awareness of organ donation.
For Matthews - who played 92 international games for Australia - it will be a return to Bendigo after he played for the Australian Cricketers Association Masters against the Bendigo District Cricket Association at the QEO last November.
Also confirmed for the game is Sydney Sixers' wicket-keeper Daniel Smith and the Melbourne Stars' Alex Keath.
And former Western Bulldogs' AFL player Paul Dimattina will line-up in the game. Dimattina played 131 games for the Bulldogs between 1995 and 2003.
