Penaluna will have scans on her right knee on Monday - the same knee that caused her to miss the best part of two years of basketball.
"We'll know more on Monday, but the first reaction is it's not looking good,'' Spirit coach Bernie Harrower said.
"Our physio is fairly sure Elyse will need a reconstruction.
"We're hoping for the best."
Penaluna's injury happened midway through the first quarter of the Spirit's dominant 86-67 win over the Rangers.
"She caught the ball and went to turn but her knee popped,'' Harrower said.
"I can't believe it. It was just one of things that didn't look bad, but the knee went on her.
"Elyse had worked really hard to get back and was starting to get back to her best form.
"She will be sorely missed if she can't go on."
The Spirit host the Melbourne Boomers at 2pm on Sunday afternoon.
The third-placed Boomers must defeat the Spirit to retain any hopes of grabbing second place off Dandenong.
Spirit guard Kelly Wilson suffered a knock to the head in Saturday night's game against Dandenong and a decision on her availability for the clash with the Boomers won't be made until just before game time.
