Petrie raced the 21.1km around the city streets in 1 hour 37.38 minutes.
The Masters champion smashed what had been the world record mark of 1 hour 42 minutes.
The record-breaking feat and gold medal capped an outstanding performance by Petrie across many days of the nine-day championships.
"To have broken the world record is an unbelievable feeling," Petrie said from near the finish line in Rosalind Park.
"This was a fantastic course to race on.
"It was well-marked, well-marshalled and there were lots of competitors."
This week's campaign was Petrie's third at Oceania Masters level.
A great run included gold in the 8km cross-country, 5km, 1500m, 800m.
She also earned a Special Recognition award for highest percentage of Masters athletes at 101.
Petrie holds the world record for the 5km at 21.34. A goal is to beat it at the Australian Masters in Hobart or the Victorian Masters in March.
The Oceania Masters finale in Bendigo included an open half-marathon which was won by Eaglehawk's Luke Preston in 1 hour 15.37 minutes.
