Martin fired a brilliant six-under par 66 at Peninsula Golf Club to lead by one shot from a group of four players.
The top 25 players at the end of four rounds earn their tour card for 2014.
Martin started his first round on the 10th hole and he got off to a flying start, making birdies on the 11th and 12th holes.
A third birdie on the par-five 17th hole saw Martin make the turn at three-under par.
Birdie on the second and third holes saw Martin grab a share of the lead.
A bogey on the par-four fourth halted Martin's momentum, but he bounced back strongly by making an eagle on the par-five fifth.
He made par on his final four holes to sign for a six-under 66.
Meanwhile, Lucas Herbert's run at the Victorian Amateur Championship has come to an end.
Herbert lost his quarter-final battle to Ben Eccles four and three at Kingston Heath Golf Club.
