The Perth left-hander will have to overcome Noppawan Lertcheewakarn, the runner-up to Dellacqua in last week's first $50,000 Pro Tour tournament, in a quarter-final on Friday.
Awaiting the winner in a semi-final could be second seed Olivia Rogowska.
Dellacqua won in straight sets on Thursday to extend her unbeaten streak on Bendigo's Nolan Street courts to 12 matches.
The third seed was too good for Japan's Michika Ozeki 6-4 7-5.
Thailand's Lertcheewakarn continued her red-hot form by dispatching sixth seed Anett Kontaveit 6-3 4-6 6-1.
In other round of 16 matches, Rogowska built on her opening round triumph with a convincing 6-2 6-3 victory against Jessica Moore.
The Melbournian will face a step up in class in the quarter-finals against Monique Adamczak.
The number eight seed rebounded from a dismal second set to defeat Sally Peers 6-3 0-6 6-2.
The other half of the draw has been blown wide-open.
Top seed Irina Falconi and 2012 champion Arina Rodionova were shock losers on Thursday.
Falconi went down after a fierce battle against rank outsider Ema Mikulcic.
The American took the first set 6-4, but Mikulcic wrestled her way back into the match.
The Croatian nabbed the second set 6-4 and held-on to upset Falconi with a 6-3 final set.
The match statistics told the story of an arm-wrestle.
Mikulcic edged points won 95-94 and converted eight of 12 break points to the American's six of eleven.
Mikulcic will face France's Irena Pavlovic in a quarter-final.
The seventh seed was too strong for the Czech Republic's Petra Krejsova in a 6-3 7-5 triumph.
Meanwhile, Tammi Patterson matched Mikulcic's surprise win in her upset of Russia's Rodionova.
Patterson was hammered 2-6 in the first set, but came back to clinch the next two sets 6-3.
The Brighton 23-year-old moves into a quarter-final clash against fifth seed Sachie Ishizu.
The Japanese star had a comfortable 6-3 6-3 triumph against Belarus' Ysaline Bonaventure on Thursday.
The order of play for Friday sees Rogowska taking on Adamczak on court one from 10am.
Over on court two, Mikulcic will face Pavlovic.
Not before 11am, Dellacqua and Lertcheewakarn will meet again on the Bendigo Tennis Association's showcourt.
On court two, Patterson will vie for a semi-final berth against Ishizu.
